Provides research and training support for NIH funded projects and projects in development, within the developmental immunology focus of the lab. Leads a segment of complex research performing in vitro and in silico experiments using human-derived tissue. A majority of effort will be dedicated towards the NIH-sponsored ECHO study, and occasional support of two additional NIH-sponsored translational studies (microPROseq and Neoimmune). Trains and supervises other lab personnel in standard immunology methods, and in cytometry and cytometry data analytic techniques. Requires experience and ability to work in a laboratory and interdisciplinary team environment with a diverse group of personnel.
Typical Assignments and Responsibilities:
Under general direction and exercising independent technical judgment, is in charge of a segment of complex research in larger projects and works independently in specialized research assignments. Analyzes and evaluates experimental data and interprets results; prepares written reports and presentations. Optimizes, performs and teaches multiple lab techniques including cell isolation, cell culture, flow cytometry, mass cytometry, novel reagent preparation, using human-derived cells and tissue. Develops, in cooperation with interdisciplinary translational research teams, protocols for specimen handling, assay performance and data capture. Performs data analysis for high-dimensional laboratory datasets. Applies software development best practices (R, MatLab, Mac OS, Flowjo, PRISM, and Windows environments) to design, implement, and test workflows and tools. Manages raw data and the associated metadata, as well as the results of data analysis workflows. Assists in regulatory compliance, personnel training, tracking of spending and documentation. Coordinates service support: repair of equipment, requisitioning of supplies and research materials and minor construction. May recommend expenditures for laboratory equipment and construction and authorize expenditures for expendable supplies.
Collaborate with Pl and Technical Associate II in the design, planning and execution of specialized research assignments related to phenotyping the immune response in humans. Work independently and collaboratively with affiliated research teams to apply novel, innovative approaches and techniques for characterizing human immune responses, with special emphasis on sample-sparing techniques with robust performance. Make scientific contributions to research projects by interpreting and evaluating observations, and based on that information, make an assessment of the scientific importance of results, and recommend additional experiments. Maintain expert knowledge in cytometry and technologies.
Optimize workflows to maintain laboratory data integrity from collection through integration. Serve as liaison with bioinformatics team to develop and execute data capture and quality control methods for laboratory data. Write, update and oversee adherence of trainees in the use of standardized procedures for maintaining data integrity.
Under PI and Technical Associate II supervision, apply existing and develop new tools for the purpose of analyzing, interpreting and preparing high-dimensional datasets. Prepare and present novel tools and findings to Pl and collaborators at laboratory meetings. Prepare summary text and graphical representations for inclusion in presentations and publications, using R, Powerpoint, Adobe Illustrator, Inkscape, PRISM and other relevant graphical tools.
Keeps abreast of trends in field of interest by studying current research literature, abstracting scientific articles of value in the prosecution of research problems and by attending lab meetings and center seminars.
Ordering and maintenance of supplies and equipment; preparation of common and new reagents (ex: custom antibody conjugation and validation for cytometry). Supervise and train other laboratory staff, graduate students, and postdoctoral fellows in relevant laboratory techniques and equipment use. Provide support and technical assistance on experimental problems that may arise during the course of their experiments.
Assures and oversees regulatory compliance for the lab and lab staff, including biosafety, chemical safety, federal and state documentation. Assure compliance with University and other regulatory policies for use of animals in research. Set up, operate, maintain and coordinate service support of complex laboratory equipment.
Bachelor's degree with major course work in the field of assignment, and minimum 3 years related experience in field of assignment.
Knowledge of BSL2 procedures, T cell culture, flow sorting, flow cytometry, mass cytometry, multiplex assays
Fluent in use of DIVA, Flowjo, Mac OS, Powerpoint, Word, Excel
Familiarity with R-package (or MatLab, PRISM)
Experience working in multidisciplinary research team
