Under the direction of Dr. Yi-Tao Yu, the Staff Scientist will undertake research in the Yu lab to improve their understanding of how ribonucleic acid (RNA) modification contributes to gene expression regulation in the cell. More specifically, she/he will lead an effort to develop a new strategy for treatment of Cystic Fibrosis (CF), which is outlined in the new grant proposal entitled " CFTR nonsense suppression by targeted pseudouridylation ".
Design experiments: 20%
Design experiments with Principal Investigator (PI) based on previous results and the literature and stay abreast of past and current literature.
Design and guide RNAs to direct CFTR mRNA pseudouridylation at the premature nonsense codon (resulting from mutations).
Design experiments to introduce the guide RNA into cells that carry the nonsense mutations. Design strategies to assay RNA pseudouridylation at the target site, and to monitor the restoration of full-length CFTR protein synthesis.
Execution of experiments: 20%
Carry out a series of experiments, including cloning, transfection, RNA isolation/detection, RNA modification, and protein isolation and detection.
Develop and install operating procedures for the laboratory, and independently operates complex equipment.
Develop and modify test methods; select procedures for data collection and handling; develops or utilizes computer programs for conversions and statistical treatment of data.
Trains, supervises and evaluates other staff and students. Directs initial training as well as continuous skill development of staff/trainees in laboratory techniques and instrumentation procedures. Counsels staff/trainees on technical problems; advises and assists staff/trainees with special research projects.
Develops and controls research project budgets in consultation with supervising faculty. Is responsible for research project budget reports and records. Recommends expenditures for major laboratory equipment and construction. Authorizes expenditures for expendable supplies and maintenance.
Fine-tuning of experiments: 15%
Experiments will not always go perfectly well. The role will entail fine tuning and repeating the experiments
Works on experimental conditions and troubleshoots.
Interpret experimental data, draw conclusions, and communicate results: 15%
After carrying out experiments, carefully collects data and analyzes them, and draws conclusions.
Communicates experimental results to the Principal Investigator (PI) and other lab members involved in the project.
Analyzes and evaluates experimental data and interprets results within the scope of the study; assesses the importance of findings in relation to the general research program.
Prepares written reports on all phases of laboratory work involved in research projects.
Contributes to the design, interpretation and final writing of research publications – thereby meeting expectations for co-authorship of scientific publications.
Participation in writing up any resulting manuscripts: 15%
Analyzes data and transposes into a format that is appropriate for publication.
Works with the PI in writing manuscripts.
Contributes to writing of research proposals, and prepares regulatory documentation required for performance of research.
Writes protocols for the conduct of research, which include: purposes of study, design of experiments, lists of materials and methods to be used and the schedule of activities.
Attend seminars/meetings: 10%
Facilitates, participates, and attends lab meetings.
Attends and presents new data at research conference(s).
Other projects and job duties as assigned. 5%
Doctoral degree with major course work in the field of assignment and a minimum of 2-5 years of relevant postdoctoral training.
Strong interpersonal and communication skills, including written communication.
