Laboratory Technician IV (S)

GENERAL PURPOSE:
Under some supervision and with latitude for exercising independent technical judgment, is responsible for carrying out complex research procedures related to protein purification, DNA cloning, mammalian and insect cell culture, and performing Luminex assays to test human and rodent sera. Plans, develops and conducts new experimental protocols. Additional work with rodent colonies, including vaccination, intransal influenza infection, euthanasia, organ dissection and primary B and T cell isolation and culture. Ability to perform and co-ordinate multiple tasks on a daily basis. Will also ensure maintenance of laboratory supplies, equipment and proper disposal of all materials.
SPECIFIC DUTIES INCLUDE:
Molecular biology work focusing on influenza and corona virology, such as gene cloning of Influenza hemagglutinin (HA) proteins, spike protein (S) of coronaviruses and HA, S protein expression and purification using baculovirus expression systems, including maintenance and infection of insect cell lines. DNA isolation using commercial maxi- and mini-prep kits. PCR amplification and sub cloning of wild type and mutant HA gene sequences into baculovirus expression vectors. May also include the culture and production of influenza viruses, and performing influenza hemagglutination (HAI) and microneutralization (MN) assays.
Isolation and storage of human and mouse serum for serological analysis; performing luminex based assays assessing antibody levels to influenza HA and coronavirus S proteins.
Independently documenting experimental protocols, as well as adapting these protocols to meet the needs of individual experiments and special situations. Collecting and analyzing scientific data. Maintaining technical expertise and awareness of trends through professional journals and research team meetings. Performing literature searches for material relevant to research projects
Operating and maintaining complex experimental equipment including Luminex and ELISA readers, spectrophotometers, PCR machines, and the like.
Involvement in mouse studies, such as assisting in the maintenance of rodent colonies, while also performing intranasal influenza infections, blood collection through submandibular bleeds and terminal exsanguination, i.m. and i.p. injections, euthanasia and spleen/lymph node harvesting.
Assist in ordering laboratory supplies as well as maintaining an accurate electronic inventory of laboratory supplies, chemicals and reagents.
Other duties as assigned.
REQUIREMENTS:
Associate's degree in appropriate discipline required.
Degree in science or engineering discipline preferred
Bachelor's degree preferred
4-5 years specialized experience in related field, or equivalent combination of education and experience required.
Molecular experience including DNA cloning and purification, PCR, protein purification and mouse work highly desirable. Experience with tissue culture, and multiplex assays desirable. Computer skills with Excel and Adobe graphics software, and/or bioinformatics experience also desirable
